Best practices for hepatic in vitro models – The practical course in Brussels, Belgium (S&T COURSE EVENT 5a)

From August 29th to September 16th , 2022, internal researchers of the Twinalt project: Edyta Wieczorek from the Nofer Institute of Occupational Medicine (NIOM) and Maxim Van den Broucke, Peldon Tenzin from the Vrije Universiteit Brussel (VUB) participated in the practical course “Best practices for hepatic in vitro models” organized on-site by VUB, Department of In Vitro Toxicology and Dermato-Cosmetology.

The three-week practical course was based on:

1) assessing the cytotoxicity of triclosan and SLS (ref) using CellTiter-Glo® Luminescent Cell Viability Assay with the use of human hepatic cell line HepG2.

2) investigating lipid accumulation induced by triclosan and Na-VPA (ref) using BODIPY 493/503 Staining of Neutral Lipid Droplets for Microscopy and Quantification by Flow Cytometry with the use of human skin stem cell-derived hepatic – hSKP-HPC.

3) determining CYP3A4 activity modulated by triclosan and rifampicin (ref) using HepaRG – P450-Glo™ CYP3A4 Assay with the use of human hepatic cell line HepaRG.

Satisfied workshop participants, in addition to the possibility of extending the basic workshop (culturing new cell lines), got acquainted with the new tests not only theoretically, but also practically.
